Detection of Anti-SS-B/La and Anti-Scl-70 Antibodies

Autoantibodies to subcellular particles SS-B/La are found in Sjogren`s syndrome with higher frequency than in SLE, and antibodies to Scl-70 are scleroderma-specific diagnostic markers. The detection method of anti-SS-B/La and anti-Scl- 70 antibod ies is double immunodiffusion technique which is the most widely employed method. The agarose plate and rabbit thymus extracts(RTE) were prepared in our own laboratory and the assay has been done with ANA human reference sera( #2, #9) supplied by AF- CDC. The results were as follows: 1) The precipitin lines were observed until 32 folds in #2 and 8 folds dilutions in #9 reference sera. This results were placed in the suggested ranges by AF- CDC. 2) The ANA profiles of the patients` sera revealed four distinct patterns. (1) Anti SS-B/La alone (2) Anti-Scl 70 alone (3) Anti-SS B/La with Anti-SS A/Ro (4) Anti SS-B/La with Anti-nRNP.
